Researchers are routinely sequencing BAC clones with 2nd Generation Technology to accompany Whole Genome Sequencing Projects performed on genomic DNA directly.
By sequencing pools of BAC clones a complex genome can be simplified giving super structure of 100 Kb to 200 Kb inserts for greatly assisting contig assembly.
The number of BAC's in each pool is driven by several factors:
-Size of haploid genome.
-Average Insert size of BAC library.
-Type of instrument your lab will be using.
-Length of sequencing reads achieved by these instruments.
-Amount of sequence data already available from the organism.
-Overall goals of project (draft coverage or standard coverage or maximum coverage).
