Nylon Filters

Screening large numbers of clones can be facilitated by robotically printing viable, bacterial cells from 96 or 384 well plates onto nylon filters. After overnight growth of the clones, the filters are processed and ultimately, the DNA from each clone is permanently fixed to a known position on the filter. These clones can be screened using standard DNA hybridization procedures. Positive clones are identified and can be retrieved from the original plates. Typically, we print 22 cm x 22 cm filters at two densities: 9,216 clones/filter or 18,432 clones/filter. All clones are printed in duplicate and we can customize the filters to meet your demands.

  5. Please indicate whether the library is stored in 96 or 384-well plates.
  6. Please indicate the order in which you would like your plates gridded if the library plates are not in sequential order.

Packaging Recommendations: 96 or 384-well Plates:

The following list is a set of recommendations for packaging your 96 or 384-well plates for shipment to our lab:

  • Stack plates on top of each other and secure them with rubber bands
  • Wrapped secured plates with bubble wrap or packing paper, or place in a box to protect plates from damage by loose dry ice pieces.
  • Place boxed (or wrapped) plates into a styrofoam container and pack the container tightly with dry ice. Note that dry ice will sublimate during shipping, so it is critical to separate your plates from the dry ice, as loose plates will get damaged during shipment.
  • Amplicon Express does not recommend use of adhesive plate seals on your plates due to the high likelihood of cross contamination when the thawed plates are opened for processing. Plates that are frozen via dry ice do not need to be sealed, though using a plastic plate cover, and stacking/rubber-banding the plates as noted above is important to your plates arriving in good condition in our laboratory.
