Shotgun Library DNA Sequencing Services

Sequencing Your BAC, YAC, or Cosmid Libraries
Shotgun Library Sequencing may be utilized after you have found a clone of interest in your
BAC or cDNA library. Briefly,
the process of shotgun sequencing involves the following steps:
- Prepare ultra-pure DNA from your BAC/YAC/Cosmid clone of interest.
- Digest or shear your ultra-pure DNA and select the appropriate size range for your library.
- Ligate your sheared DNA into the vector of your choice.
- Transform the vector + insert into a suitable host, typically a commercially available brand of modified E.coli.
- Plate the transformation onto growth media and allow the clones to express a selection mechanism to indicate
the presence of clones with an insert and clones without an insert.
- Pick the positive-insert clones into a 96 or 384 well format for arraying and storage.
- Grow the selected plates of clones for sequencing, and prepare highly pure DNA.
- Perform sequencing on the highly pure DNA.
- Analyze, assemble, and annotate your sequence data.

Whole Genome DNA Sequencing Services

Complete Sequencing & Assembly for Your Whole Genome
Whole Genome Sequencing is similar to the shotgun library sequencing process, though the
library itself is prepared from the DNA of an entire organism instead of an individual clone. Whole genome
sequencing is typically successful with organisms having smaller total genome sizes. Briefly, the whole
genome sequencing process occurs as follows:
- Prepare ultra-pure DNA from your organism of interest.
- Digest or shear your ultra-pure DNA and select the appropriate size range for your library.
- Ligate your sheared DNA into the vector of your choice.
- Transform the vector + insert into a suitable host, typically a commercially available brand of modified E.coli.
- Plate the transformation onto growth media and allow the clones to express a selection mechanism to indicate

BAC End Sequencing is a method of choice for proving overlap of two clones in
a contig map that has been generated by BAC Fingerprinting techniques.
End sequences can be aligned to find matching regions, and confirmation of a physical genome map is made

Genewalking Services
|
Close Assembly Gaps and Completely Sequence Your Gene of Interest
Amplicon Express Genewalking Services are very useful for the researcher with a clone of
interest that is not large enough for a shotgun library, and is small enough to walk in a reasonable
amount of time. Genewalking involves the process of sequencing the ends of a clone, and designing a new
primer set on the 3' ends of each read in order that sequence generation can continue into the unknown
frontier of the selected clone. The rate at which a genewalk can be completed typically depends on the
speed of primer synthesis, and though it is one of the slower methods of sequencing, it is still very
useful for closing gaps in genome assemblies and confirming a clone that may be used later for protein
expression or other purposes.
